androst-4-ene-3,17-dione(AD)is the intermediate of hormone medication,which is impossible replace by others.It plays important effcetion of adjusrment to the organism. All the hormone medication are produced from the material of AD,so to speak.At present,the production of AD is transformed from the chemic extraction from yam amole to fermentation by microorganism.The fermentation is,high yield,facile process,cost low,and is not restrict by material,so that the produce is easy on a large scale.So we need explore anvanced technique to produce AD.The article quested for some studies on producing AD by fermentation,and finishined the following jobs.Established a series of methods of analysing AD,which are fast and effective.Just like TLC,Ultraviolet absorption spetrometry,HPLC.Supplied the predition to the following experiment.Solved the indissolubility of Phytosterol.The Phytosterol was treat with adding alcohol, cyclodextrin,Tween80 and ultrasonic.Employing Tween80 to dieperse substrate is the best method.Optimized fermentation medium in culture flask was determined Through The single factor tests and multiple factor orthogonal tests,fermentation medium in culture flask was determined,which was composed of 20g/L glucose,20g/L peptone,0.8g/L K2HPO4,0. 7g/L,MgSO4·7H2O,9g/LCaCO3.Optimized culture conditions were determined.The growth curve of bacteria in seed medium was made,50mL fermentation medium in 300ml flask can obtain the highest AD output,and optimum pH value for AD accumulation was 7.0,optimum temperature we for AD accumulation was28℃,6%was selected as the best amount of inoculation,250r/min was the best rotate speed,0h was the besttime for adding Phytosterol.Optimized culture conditions were determined on the level of fermentor.The ideal fermentation condition for AD accumulation is that PH value is 7.0.and temperature is 28℃.Further more,this paper screen the best tactics of stiring and aeration,which is keeping the aeration volume 5L/min through the whole course of fermentation,while regulating different stiring speed in the course of fermentation:keeping the low speed of stiring 300r/min at the first of the course,When the DOT has decreased,changing the stiring speed into 600r/min.The single batch-feeding fermentation was adopted in this paper,and we fumble the different method of feeding,finding that the exponential feeding method is the best way for the output of AD.
