Pregabalin is a new medicine to cure epilepsy and neuralgia, the good curative effect of pregabalin brings hope to many epileptics, and it has real good commercial future. Nowadays there is less disquisition about synthesis of pregabalin in China, so this project has very good developing potential.Based on theoretical analysis, this project focuses on the experiment process in the resolution 3-(Carbamoylmethyl)-5-methyl-hexanoic acid with (R)-(+)-α-phenylethylamine, and then improve the method of experiment by analysis of resolution product purity. The project finds a new system of resolution by theoretical analysis , and analyzes influence of product purity of different conditions in reaction.On the basis of the above experiment, take out the research and improve other key courses of the reaction of the Pregabalin, raise the new technology and ideas, through the improvement and replacement of the reaction, the reaction processes, and the catalyst, improve the process, improve the yield of reaction, so as to cut down the costs of the production and raise the efficiency of the production. Such as the recycling process of the rich R-isomer, consider the hydrolysis of raw materials, anhydride ring and ring-opening reactions on the details of the influence of various factors,so as to improve the recovery process of the Heterogeneous systems, and to cut down the costs and save the energy. Also, take the study of the mechanism in the degradation of the amide of the Hoffman reaction in accordance with the relevant patent described, and introduce a suitable catalyst to improve the yield of Hoffman degradation.
