Xylo-oligosaccharides,as one kind of most effectively functional oligosaccharides,could be used to promote the proliferation of Bifidobacterium in the intestinum.This paper presented an application of response surface methodologies for production of xylo-oligosaccharides from bagasse by sulfuric acid,sodium hydroxide and hydrogen peroxide with microwave.The results were listed as follows:1.Xylo-oligosaccharides were prepared from bagasse with enzymatic hydrolysis by microwave.Results indicated that when the bagasse were heated by microwave at 0.9MPa for 17min and subsequent hydrolysed by 0.7% xylanases for 24h,the content of reducing sugar reached 9.21mg/mL.2.The TLC demonstrated that xylobiose,xylotriose and xylotetrose were the main compositions in the hydrolysis of microwave.There was also a little of xylose. 3.The XRD indicated that cellulose crystallization of bagasse had not been destroyed.X-ray diffraction mountain intensity of bagasse after microwave digestion was stronger than bagasse samples.There was more xylan being dissolved.4.The purification of xylo-oligosaccharides was studied.The decolorized xylo-oligosaccharides liquid with xylobiose,xylotriose and xylotetrose as major component was obtained with three steps:separating the hydrolysis solution with an activated charcoal column,elution with water and 30%ethanol.94.3% of pigment was removed.The elution liquid of water TLC demonstrated that pigment and high polymer boilings were the main compositions in the hydrolysis of microwave.The elution liquid of ethanol TLC demonstrated that xylobiose,xylotriose and xylotetrose were the main compositions in the hydrolysis of microwave.There was also a little of xylose.5.Mensurating the content of xylo-oligosaccharides by HPLC.The results were listed as follows:the content of xylo-oligosaccharides in the xylo-oligosaccharides solution was 50.9%,the content of xylobiose,xylotriose and xylotetrose were 48.9%,the content of monosaccharides were 9.7%.
