In this paper,Zebrafish embryos and SH-SY5Y cell were used as test models,and a series of toxicity test were established.A method of chiral separation of four enantiomers of beta-cypermethrin was developed by high performance liquid chromatography(HPLC).The thesis aims to evaluate toxicity of beta-cypermethrin and its four enantiomers to zebrafish embryos and SH-SY5Y cell,and also provides the evaluation standard for environmental organisms.A method for chiral separation of four enantiomers of betacypermethrin was developed by HPLC on the CHIRALCEL OD and CHIRALPAK AD columns.The effects of different proportion of mobile phase(on OD and AD columns)and column temperature(on OD column) were studied.Besides,assignment of absolute configurations to the resolved four peaks was made by comparing chromatogram from previous study.According to the OECD standard,the effects of beta-cypermethrin and its four enantiomers to zebrafish embryos were researched.At 48-h,beta- cypermethrin and its four enantiomers had the greatest effect on the hatching rate of the zebrafish embryos.The most embryotoxic isomer is 1R-cis-αS,next is racemate and 1R-trans-αS.Embryos exposed to these isomers showed various developmental abnormalities(yolk sac edema, pericardial edema and crooked tail),and crooked tail was the most sensitive endpoints.On the other hand,no effect on morphology was recorded for both 1S-cis-αR isomer and 1S-trans-αR isomer.Both of the degree of edema and crooking increased with increasing concentration at 72-h.The test evalutes the cytoxicity of both racemate and four enantiomers of beta-cypermethrin on neuronal cell line SH-SY5Y.According to the results,all of the five forms of beta-cypermethrin showed dose-dependent effect.Cells exposed to racemate and 1R-cis-αS isomer showed the greatest differentia(1.52 times)at the concentration of 100μmol/L.1R-cis-αS isomer has been shown to be the most cytotoxic compound,and the toxicity order of the enantiomers was 1R-cis-αS>1S-cis-αR,1S-trans-αR>1R-trans-αS.
