In this paper,the extraction,isolation,purification,structure and bioactivities of the total flavonoids from Water Hyacinth(Eichhornia crassipes)was studied.Single-factor experiments of extracting total flavonoids from Eichhornia crassipes were separately carried out using of hot water soaking,ultrasound and microwave extraction methods. The optimum conditions of extracting total flavonoids from Eichhornia crassipes were determined through orthogonal experiments which were on the basis of single-factor experiments with microwave extraction method.The optimum conditions were: microwave time-2min,ethanol concentration-50%,feed ratio-1:80,microwave firepower-M.Low.After degreasing,disposition of ethanol,the WCTF was applied to AB-8 column.Then we got a purified total flavonoids fraction WCTFS.The purity of WCTFS was confirmed by HPLC.The chemical structure was analyzed by TLC,UV and so on.The results showed that WCTFS contains flavonoids,mainly flavonols.The effects on scavenging·OH and·O2-of Eichhornia crassipes total flavonoids were studied by Fenton system and Pyorgallol Autoxidation method respectively,which were compared with the effect of manitol and Vc.After mixing WCTF,WCTFS with Aureobacidiumpullulans,Aspergillum niger,Chaetomium globosum,Penicillium funiculosum respectively for 24 hours and 48 hours,the results of hatching compared with the results of sterile water showed that both WCTF and WCTFS have well bacteriostasis effects,WCTFS was better than WCTF.The bacteriostasis effects of WCTF and WCTFS were studied on bacteriostasis circles experiments of Escherichia coli and Staphylococcus aureus.The results showed that both WCTF and WCTFS do not have bacteriostasis effects when their concentration are 0.01mg/mL.
