Polyphosphatester is a biodegradable material which has been aroused great interests in recent years. Because of the similarity to biomacromolecules like nucleic acids, polyphosphatester could be hydrolyzed or enzymically decomposed in physiological conditions, and thus has good biocompatibility and biodegradable capacity and can be used as medical polymer materials. In this paper, we prepared a novel cross-linked structure contained amino phosphoric esters based on poly(propylene fumarate) (PPF), which has been extensively explored at present. Its drug release behavior in vitro was also investigated.Firstly, PPF was synthesized using condensation polymerization reaction of bis (1, 2-propyleneglycol) fumarate (BPGF) and characterized by FTIR, HNMR and GPC. The effects of reaction temperature, reaction time and other conditions on polymer synthesis were studied. The results showed that at 180℃for 10 hours, PPF can get the molecular weight up to 23813.MCPBA was used as an oxidant to epoxide some double bond in the main chain of PPF and the final epoxy product was characterized. Poly-phosphate fumarate cross-linked copolymer was synthesized through cross-linking reaction with diethyl aminomethylphosphonate.The release profile in vitro of 5-FU charged in poly-phosphate fumarate cross linked copolymer was studied. The cumulative drug release was 35% in vitro in 50h, which indicates a sustained release behavior of poly-phosphate fumarate cross-linked copolymer.
