Chitosan is a kind of abundant, reproducible and natural polymer with non-toxicity, biocompatibility and biodegradability. But the limitation of its dissolubility confined its application. Chitosan should be modified to settle these issues.A new kind of chitosan-based amphiphilic has been synthesized by the reaction of N,N-dialkyl chitosan and Maleic anhydrides in homophase using THF as solvent, N,N-dialkyl chitosan was synthesized using an initiate method contrived by our lab. The structure of products have been characterized and testified using Fourier transform infrared (FTIR), EA and 1HNMR which show that we have successfully synthesized these new chitosan-based amphiphilic derivatives. The dissolubility experiment found that O-butylene diacid ester-N,N-dialkylated chitosan could be dissolved in Tetrahydrofuran, Pyridine and N,N-dimethyl formamide, but could not be dissolved in acetone and cyclohexane.In this work, the properties of O-diacid mono ester-N,N-dialkyl chitosan monolayers have been investigated. Result showed that with decreasing MW of chitosan backbone, increasing of alkyl chain and different of the hydrophilic groups, collapse pressure of chitosan-based derivative monolayer was enhanced, and the static surface elasticity was subdued.In the drug release experiments, vitamin B12 is used as a model drug for this study in the paper. The experimental results indicate that these new chitosan derivatives can form nanoparticles, the drug-releasing rates of vesicles made from longer alkyl chain and larger molecular weight of chitosan are slower than the others, and drug-releasing equilibrium amount as well.The germ resistance of this series of chitosan-based amphiphilic derivatives had been studied. The experimental results indicated that the germ resistance of these derivatives had good antibiotic ability. The antimicrobial activities changed with different chitosan molecular, alkyl chains and hydrophilic group of these chitosan derivatives.
