| The cross-linked chitosan were firstly prepared by glutaraldehyde crosslinker, then Crosslinked chitosan-hydroxy proline resin(HCCR) was synthesized as substrates of CCR and L-hydroxy proline(Hyp) by dicyclohexyl carbondiiminde (DCC). The optimum technique parameters were such as below: mass ratio of CCR:Hyp is 1:1, molar of DCC:Hyp was 1.5:1, pH is 6, reaction time 10min, the grafting ratio of Hyp reached to 80.38%. Both IR and 13C-NMR prove that amide linkage is formed between CCR and Hyp. The scanning electron microscope shows that HCCR are porous microspheres, its average diameter and pore diameter are respectively 700~1000 and 50~100μm. The HCCR's dissolution ratio in 5% HCl and 5% NaOH are 0.02% and 0.03% respectively, the water content is 91% and stacking density of dry resin is 0.096g/mL.The equilibrium adsorption amount of Hyp by HCCR is 75mg/g at the pH being 3, the initial concentration 2.5mg/mL and adsorption time 30 min. The dynamic exchange capacity of Hyp, Gly, Phe and Pro are 70, 10, 13 and 27mg/g respectively at the adsorption velocity of flow 3mL/min. Hyp adsorbed is eluted from HCCR with 70% ethanol solution whose pH is 9 and elution flow rate is 2mL/mim, and washout ratio reaches 93.62%. The Hyp is separated effectively with Gly, Phe and Pro by controlling the velocity of flow. |
