The study preparation Bifidobacterium bifidum freeze-dried powder and microcapsules to resolve the question of B.bifidum product in deposit and distribution.Preparation B.bifidum freeze-dried powder and microcapsules of high amount of viable,then researched the characters of B.bifidum freeze-dried powder and B.bifidum microcapsules.Fist of all,three kinds of preparation technology were researched to increase the viable count of B.bifidum freeze-dried powder.The methods were as follows,freeze-dry after using the medium of milk fermentation B.bifidum,freeze-dry with mannitol and skim milk powder as cryoprotectants after using the PTYG medium culture B.bifidum,and using the gelatin and skim milk powder as cryoprotectants.The viable count of B.bifidum was large for the second one,it can reach 1.31×1010 cfu·g-1,and it was stable in gastrointestinal environment.The second one was tested for the industrial production,and the preparation technology was obtained.The result showed the highest amount of viable B.bifidum in freeze-dried powders could reach 1.67×1010 cfu·g-1.The viable count of B.bifidum could keep 5.83×109 cfu·g-1,when freeze-dried powders were disposed in gastrointestinal environment.The other characters of B.bifidum freeze-dried powder was fine.Classic accelerating test showed that the B.bifidum freeze-dried powder had better stability of storage.More than 108 cfu·g-1 bacteria remained alive after one year storage for room temperature.In addition,we research the extrusion,emulsion,polymerization method of B.bifidum microcapsules.The viable count of B.bifidum was large for the emulsion 2,that alginate sodium,calcium chloride and chitosan were used for the principal wall material in production. The result showed the particles were 10~20μm,the amount of active B.bifidum were more than 3.01×109 cfu·g-1 and the embed ratio could reach 90.36%.When microcapsules were disposed with artificial gastric solution and bile solution,the amounts of viable B.bifidum in microcapsules were more than 109 cfu·g-1.It indicated it had the high acid tolerance.When disposed with artificial intestinal solution for 35 min,all microcapsules were nearly disintegrated,and the release ratio reached 90.93%.The other characters of B.bifidum freeze-dried powder was fine.Classic accelerating test showed that the B.bifidum microcapsules had better stability of storage.More than 8.61×108 cfu·g-1 bacteria remained alive after one year storage for room temperature.B.bifidum microcapsules compared to B.bifidum freeze-dried powder,the characters of microcapsules were superior to the freeze-dried powder.Particularly in resist acid,bile acid-resistant, storage stability,and other aspects,the B.bifidum microcapsules performance more obvious advantage than freeze-dried powder.B.bifidum microcapsules were disposed in gastrointestinal environment,the ratio of amount of active B.bifidum was 60.13%,and the B.bifidum freeze-dried powder is 34.91%.Classic accelerating test showed that the B.bifidum microcapsules had better stability of storage,the ratio of amount of active B.bifidum was 28.60%,and the B.bifidum freeze-dried powder only is 1.37%.
