Petroleum hydrocarbons is one of the main pollutants in marine environment. In this paper, sea urchm(Hemicentrotus Pulcherrimus )were selected as test organism. Experimental research on the sea urchin genome-wide DNA methylation level and the probability of complement C3 gene mutation polluted with the petroleum hydrocarbons. Investigating the toxic effects of petroleum hydrocarbons on the marine organisms in molecular level. At last, probing the toxicity mechanism of petroleum hydrocarbon pollution on aquatic organisms. So it can provide reference to early warning and risk assessment for petroleum hydrocarbon pollution on the marine environment.Analyzing chemical substances on biological DNA methylation is an important molecular toxicology means to assess the level of its biological toxicity. In this paper, high-performance liquid chromatography is used to analyze the sea urchin DNA methylation changes which exposed to different concentrations and time of No.0 dispersed diesel oil. The experiments result shows that petroleum hydrocarbons can cause sea urchin genome changes in the overall level of methylation, with the extension of exposure time, there would be a low methylation phenomenon, but we haven't found significantly dose-response relationship, which indicate that petroleum hydrocarbons can impact sea urchin DNA methylation and its regulation in vivo.The sea urchin complement system is an important function that response body immune capacity. In this paper, In order to explore that if the 5-methyl cytosine is "hot spots" of mutation, genetic indicators complement C3 gene mutation was further studied on the basis of the study of indicators of sea urchin epigenetic DNA methylation exposure to low concentration petroleum hydrocarbons. The test results found that one primers (primers B-6) of the PCR product exist the polymorphism, 1184 occurred in the conversion of C-T mutation, that Amino acid from threonine to isoleucine. And it was also found as the concentration of petroleum and exposure time increased, the probability of mutation of sea urchin complement C3 gene increased too. It is indicated that petroleum hydrocarbon pollution can cause DNA mutations in sea urchin.This paper analyze the DNA methylation content and complement C3 gene mutation rate of the sea urchin after polluted by petroleum hydrocarbon. The experiments results found that the degree of DNA methylation reduced but the rate of complement C3 gene mutation increased as the exposure time extend of the high exposure concentrations Group. It is suggested that the existence of the 5-methyl cytosine may not be "hot spots "of gene mutation .
