Content:Nucleosides are the most frequently used for the treatment of herpes,AIDS,and viral hepatitis,their target are the reverse transcriptase and DNA polymerase of RNA viruses.Their similar structures have competed to the centre of enzyme,inserted to the viral chain and made an end of it,at last,restrained the copy of viruses. Currently,with over 20 drugs currently approved for the treatment of viral diseases(above 50%) and a number of candidate in the clinical trials.However,nucleosides also have contributed to the aspect of antiviral and antitumor,and nowadays so many nucleosides already were launched in the market,but that is not enough for requirement. Many scientists over the world are interested in the research of the antiviral drug.In recent years,new and emerging viruses,such as new strains of hepatitis and herpes viruses,Ebola,West Nile,SARS,and the flu which caused by H1 virus in Hong Kong this February,have shown their lethal potential.Furthermore,the threat that viruses and other microorganisms could be used as biological weapons in warfare or bioterrorism has brought antiviral research to the forefront.Although vaccination is a valuable preventative tool for certain viral infections,new and effective antiviral agents are needed to prevent acute and chronic viral infections.Nucleosides common character is every compound all formed by sugar and base,in the matter of chemical constitution,nucleosides and natural nucleosides have very similar character.So we can change nucleosides from two parts,from sugar and from base.All these changes are good for antivirus activities,but sugar part is very important for it,and changing sugar part can increase antivirus activities,at the same time,it can also decrease side effects.In view of these reasons,my thesis is consists of design,synthesis and analysis of 3'-methylolnucleosides and 4-selenosugar. In addition,we also present the class of nucleosides.
