Now, people put more attentions on their life quality due to extremly abundance of merchandises. Scientific research shows that natural astaxanthin with remarkable anti-oxidant and anti-aging capacity will let people keep away from the cancer and improve immunity.In this dissertation, the astaxanthin was successfully extracted from Hawaii Haematococcus pluvialis by the extraction method using supercritical carbon dioxide fluid. The two products astaxanthin and a unknown material from the effective components were obtained by concentrating , saponificating and column chromatography technology. The two products were purified by recrystallization repeatly. The dark red product was identified as astaxanthin, and another product as stigmasterol by FT-IR and UV-Vis.The carotenoid quantity in the extracted products were determined by using UV spectra. Besides astaxanthin, other kinds of carotenoids also existed in the extracts. Therefore, the quantity of astaxanthin is increased with the increasing of carotenoids in the UV spectra. In the experiment, the conditions of the maximum quantity of astaxanthin contained in the extracts were selected as the conditions of supercritical carbon dioxide fluid extraction process. The two products were characterized by UV and FT-IR, and the content were determined by HPLC.The optimal conditions extrating astaxanthin by supercritical carbon dioxide fluid were following, the work pressure of extration was 35 MPa, the extration temperature was 35℃, entrainer was used by methanol and acetone (1:1, v/v). Separation pressure and temperature were 5Mpa, 25℃, respectively. The 35 Mpa selected as work-pressure was due to the limit of the supercritical carbon dioxide extraction equipment. The inflexion temperature (35℃) was selected as operation temperature because the recovery ratio of astaxanthin firstly increased, then decreasing with increasing of the temperature.
