The object of this article was to obtain the mutagensis strain which was high yield of ergot alkaloids.The mycelium of Claviceps paspali was treated by lywallzyme to obtain the protoplast. The protoplast which was treated by UV-irradiation and the different concentrations of the nitrosoguanidine grown in the regeneration medium. The stable mutants with increased titers were obtained by the lightness of fluorescence and the fermentation.The productivity of ergot alkaloids was measured by the way of Van-Urk . The results showed that the productivity of ergot alkaloids was increased by 16 times, from the initial titers 90 mg / L to 1600 mg / L and the unit productivity of ergot alkaloids was increased by 17 times, from the initial titers 22.5 mg / L/g to 400 mg / L/g after continual transfers. The results indicated that the UV-irradiation and the protoplast nitrosoguanidine mutagenesis were feasible approaches to increase the productivity of lysergic amide of the C.papspali strain.The condition of the fermentation of Claviceps paspali was optimized. The content of tryptophan and phosphate could effect the productivity of ergot alkaloids . The different inoculum size and temperature could effect the amount of ergot alkaloids. The amount of ergot alkaloids could reach the top when the content of tryptophan was 0.08%. The content of phosphate could produce bad effection to the productivity of ergot alkaloids. The period of the fermentation can be shorted when the inoculum size was 20%.Ergot alkaloids was distilled from the fermentation liquor of Claviceps paspali by the macroporous resin of D101.The single ergot was obtained by the way of TLc and column chromatography. The results indicated that macroporous resin D101 could absorb the ergot alkaloids effectually and 10.2g ergot alkaloids could be obtained from 7L fermentation liquor.
