Due to the increasing of vegetable crop area in our country, especially, extending and using greenhouse in large area, single crop was planted continuously for many years, damage of root-knot nematode was increasingly serious, the yield reduction rates was 20-30%, and even 100% in serious case. Root-knot which were damaged by nematode was collected in greenhouse in Hexi area ,Gansu province, in June to September 2007.The root-knot nematode was separated and identificated at laboratory, toxicity test that root-knot nematode suffered from single agents and compounded agents had been done,the main result were showed as follows:1,The main morphology of nematode were showed: body length of female was 599.0 (510.0-690.0)μm, width was 439.0(409.0-473.0)μm, length of stylet was 15.4(15.0- 16.0)μm. Distance from dorsal esophageal gland orifice to stylet knobs was 3.6(2.0-4.3)μm, female nematode was pyriform, excretory pore was on stylet knobs. Dorsal arch of perineal pattern was high. Striaes shaped was form smooth to wavy, lateral lines indistinct.According to these morphology of nematode,pathogen was identified as Meloidogyne.incognita in Hexi area ,Gansu province.2,Toxicity test of single agents and compounded agents to M.nematode. Nematode was soaked by pesticide solution for 24h, LC50 of 8 pesticides(fosthiazate,phoxim, Abamectin etc.) were measured: Phoxim 16.2127μg/ml,dazomet 16.8900μg/ml, carbosulfan 3.7800μg/ml,Ethoprop 8.6987μg/ml,chlorpyrifos 1.7809μg/ml,aldicarb 2.8922μg/ml,Fosthiazate 12.8143μg/ml, Avermectins 1.5912μg/ml;Toxicity of compounded agents were measured: when Avermectin / phoxim=3/8, the most toxicity of mixture was 0.1838μg/ml;Avermectin / carbosulfan=3/8,the most toxicity of mixture was 0.4623μg/ml;Avermectin /Chlorpyrifos =2/9,toxicity was1.3599μg/ml; Aldicarb/chlorpyrifos=11/2,toxicity was2.0474μg/ml. Synergism of the six mixtures was obvious.Considerring toxicity and other facters comprehensively, the portfolio(Avermectin/ phoxim=3/8) was used to manufacture granules,finally.3,Definition of granule formula & product efficacy test. The screening sand was used as carrier, the diatomite as adsorbent, and best bonding agent was selected from Polyvinyl alcohol,β-cyclodextrin,polyvinyl acetate,paraffin, added proper alertness colors. Finally granule formulations was defined.The result of efficacy test showed that, nematode decresae rate reached 95.45% with application dosage of 18.75kg/hm2 after 30 days.Efficacy granules showed that:nematode population decrease reached 95.45% after suffered drug 30 days ,significant difference from pharmacy control.
