Coix lachryma-jobi kernel has rich nutrition and special health value. The soluble polysaccharide called coixan haven been proven to have several bioactivities, including anti-tumor activity, hypoglycemic action. In this paper, Coixan were extracted and purified. The purity of molecular weights of coixan were analyzed. At the same time, the brewing technology of kelp and Coix lachryma-jobi kernel was investigated.(1) Two different methods, traditional hot water extraction and ultrasound extraction were applied to separate coixan from Coix lachryma-jobi kernel. The obtained optimal condition of hot water extraction was as follows: extraction temperature 95℃, extraction time 4h, solid to liquid ratio 1:25, pH=5.2, the yield of polysaccharide was 1.25%. The obtained optimal condition of ultrasound extraction was as follows:ultrasound extraction time 45 min, ultrasound power 200W, extraction temperature 85℃, and solid to liquid ratio 1:25, the yield of polysaccharide was 1.41%. In contrast to other extraction methods, the ultrasound technique could be a better method for extracting polysaccharides from Coix lacryma - jobi kernel.(2) Sevag, enzymatic hydrolysis and TCA were used to remove protein from the polysaccharides. The results showed that with concentration of papain 0.14%, polysaccharide solution was enzymatic hydrolysis for 2 hours in 55℃water and pH6.5. The removal percentage of protein was 68.5%, and the loss of polysaccharide was 9%.(3) The coixan was isolated and purified by Sephadex-75, the pure coixan was identified by Waters HPLC, which did not contain protein. According to the analysis of IR fingerprint characteristics absorption, the result showed that the configuration of coixan wasβ–glycosidic.(4) Refined coixan consisted of rhamonse , arabinose , mannose , xylose, galactose and glucose with an estimated molecular weight of 1.49×104。(5) Using kelp and Coix lachryma-jobi kernel as the main raw materials to brew the health wine. The obtained optimal condition of kelp hydrolysis,the ratio of materials,yeast amount were determined by experimrnts. The heath wine that had style of raditional rice wine and kelp was rich in the nutrient composition and low-alcohol. The wine meet the demand for China's health wine market and had good market prospects.
