The aim of this study is to optimize the Broad Bean protein hydrolysis condition, studied Broad Bean protein hydrolysates (BPH) bioactivity that promoted Lactobacillus bulgaricus growth in MRS medium. Preliminary studied the mechanism of BPH promoted the Lactobacillus bulgaricus growth in MRS medium and separation the bioactivity peptide.First, we determined the extraction process parameter of Broad Bean protein: liquid ratio of 1:12, extraction pH value is 8.0, ultrasonic power is 660 W, extracting 20 min, Broad Bean isolated protein contained 89.32% protein.Second, we optimized the Broad Bean protein hydrolysis condition: heat treatment broad bean protein 15 min under 90℃, the reaction temperature is 50℃, pH value is 8.5, enzyme and substrate ratio is 96000 U/g, substrate concentration is 6%, the hydrolysis degree reached 15.55%.Thirdly, we studied the effect of BPH on Lactobacillus bulgaricus growth in MRS medium. With the 2g/L concentration BPH added to the MRS medium, the promoting the growth of Lactobacillus bulgaricus is most obvious, BPH increased counts of Lactobacillus bulgaricus by 1.0 log cycles after 20h incubation at 37℃when compared with unsupplemented MRS. The BPH played a catalytic agent of peptide transport system role to promote the growth of Lactobacillus bulgaricus.Finally, the bioactivity peptide was isolated from Broad Bean protein hydrolysates by Sephadex G-25 gel filtration and RP-HPLC.
