Nowadays the biological and medical diagnosis requires more advanced analytical systems.When it comes to this problem,Microfluidics and Nanofluidics appear to be promising analytical tools.In my works,they are about preconcentrating and separating biological and medical molecules using micro- and nano-devices.Daunorubicin(DAU) and Doxorubicin(DOX) were separated by cross-shape microchannels.FITC-BSA and FITC were preconcentrated by hybrid miro- and nanochannels.It would be fantastic to combine the preconcentration part with separation part to analyze the biological and medical molecules of low concentration.In the first chapter,various fabrication methods of hybrid micro- and nanochannels were introduced.And the reports on the preconcentration techniques of microfluidics and nanofluidics as well as the electrophoresis techniques of nanofluidics were reviewed.In the second chanpter,CE-LIF was employed for the separation of anti-tumor medicine. The DAU and DOX are naturally fluorescent which were favorable to the experiments. Adding 40%acetonitrile to the borate running buffer significantly improved the separation efficiency and the fluorescence detection sensitivity.DAU and DOX were separated successfully in the human serum solution.In the third chapter,the double lithography and double chemical wet etching method was used to fabricate the hybrid micro- and nano-devices.These devices were applied to the preconcentration of FITC-BSA and FITC,since the overlap of the double electrical layers. FITC-BSA was preconcentrated using the depletion region,while the FITC was preconcentrated in two ways including the way usually to preconcentrate small molecules and the way similar to preconcentrating proteins.This paved the way to the integration of the preconcentration and separation of medicines.
