An efficient anaerobic reactor with an effective volume of 20L has been designed, according to the characteristics of anaerobic pecked bed reactor. Pen manure to as raw material for fermentation, the raw water CODcr was adjusted between 4 820 and 54 000 mg/L the whole process was 55 days which has been divided into two stages according to the raw of hydraulic retention time:the prophase (44d), the disposal concentration of reactor was directly advanced by increasing the raw water concentration, the hydraulic remain time (HRT) was maintained 2 days,7~8 days per cycle; the anaphase (11d), the disposal concentration of reactor was indirectly advanced by shortening the residence time, and the influent concentration was unchanged,2~3 days per cycle. CODcr, pH, Volatile components and the content of TN, Tp and NH3-N were determined through to the whole procession in the anaerobic treatment system, and analysis of operating characteristics of the system.Results demonstrated that:In the fermentation section 1~55d, the whole system could be stably operated, with a stable CODcr removal efficiency which were between 67% and 88%; and high capacity in removing TN,Tp and NH3-N. The fermentation process was divided into every stages according to hydraulic retention time and the raw water CODcr, the chang of nutrition index of fermented liquid in the fermented course was monitored at each stage. The results showed that while the environment temperature conditions was controlled, the pH of fermented liquid changed within the specific limit,but did not change much wholly; through the whole fermentation, the nutrition index of fermented liquid was added with the increase of the raw water CODcr. After the process of treatment, Biogas slurry fertilizer can be used as organic fertilizer application in agriculture. In order to investigate the microbial community structures and dynamic drift in different stages of anaerobic pecked-bed reactor treating pig manure, total bacterial genomic DNA was extracted from anaerobic sludge and produced water in anaerobic pecked-bed reactor. The 16S rDNA genes were amplified with two universal primers (F357 and R517). These amplified DNA fragments were separated by DGGE as to get microorganism community DNA characteristic fingerprint. Furthermore,genomic sequencing and the sequences were used to constructed phylogenetic tree. And cloning analysis of microbial community structure in influent. Results indicate that each stage of APBR owns its specific ecological community, Influent water has a significant impact on total bacterial community structures. Microbial community structure of Anaerobic sludge is basic steady, but the specific bands in sludge samples indicated that microbial communities in different periods had specific and common ecological community, demonstrated that bacterial succession correlated with the system load; Microbial populations diversity of produced water was decreaseing with the process of anaerobic treatment.
