Onion Oil Extraction Process And Optimization

Onion is a traditional medical & edible dual-use plant with glorious cultivating history. Onion has a relatively high content of functional substance, especially the sulfur-containing compounds like allicin, which was produced by the hydrolyzing of alliin via alliinase. In the past several decades, onion has attracted more and more attention because of the bioactivity research of the sulphur-containing compounds. This study was to extract sulfur-containing compounds and onion oil from fresh onion and determine the content of thiosulfinates in it. As one of the most important bioactive thiosulfinates in onion, allicin was selected as the index of the thiosulfinates content in the extractant.The onions are dropped in hot water of 70℃for 60 min to make the alliinase inactivation, then ultrasound-associated extract for 60 min at least, followed enzymolysis for 20 min, and use CH2Cl2 to extract, the yield is higher. CH2Cl2 was the best among the solvents tested during the experiment. Orthogonal experiment L9(34) was introduced to optimize the extraction procedure. The top extraction efficiency was obtained at the condition of 65℃and 70min for enzymic inactivation,70min for ultrasound-associated extraction, followed enzymolysis for 25 min, and use CH2Cl2 to extract. Under these conditions the extraction efficiency of onion oil reached 0.138g per 100g fresh onion,5.85mg thiosulfinates per 100g fresh onion. The content of thiosulfinates in onion oil arrived at 4.397g per 100g.
