The 1,5-benzothiazepines are a kind of seven-membered ring heterocyclic compounds which possess many physiological properties. They are often used as tranquilizer and medicines for cardiovascular disease. Scientist shown great ardor in it's synthesis, structure, and physiological properties research. Oxadiazoles are naturally occurring five-membered heterocycles with utility in synthetic and medicinal chemistry. They serve as the core components of a large number of substances that possess a wide range of interesting biological activities. The 1,2,4-oxadiazole scaffolds are associated with significant biological activities, such as anti-hypertensive, antimicrobial and anti-inflammatory . Furthermore, 1,2,4-triazole derivatives have been reported to exhibit antifungal, anti-inflammatory and antimicrobial activity . The C=N imine double bond in the molecules is one of the major parts where chemical reactions occurred. Consequently, we combined 1,2,4-oxodiazole and 1,2,4-triazole which have strong biological activities with 1,5-benzothiazepines by 1,3-dipolar cycoladdition reaction respectively, and a series of derivatives with novel structures were synthesized.The thesis includes two major parts:The first part is about literature review:The first part is the literature review of 1,5-benzothiazepines. The important synthetic methods, structure and chemical transformation of l,5-benzothiazepine derivatives that have been studied in China and other countries were explicated systematically.The second part is about experimental content. Major works as follows:1. Synthesis of new 1,2,4-oxadiazolo[5,4-d][1,5]benzothiazepine derivatives.A new series of 1,5-benzothiazepine derivatives containing 1,2,4-oxadiazole were synthesized from 1,5-benzothiazepines and ethyl chlorooximidoacetate via 1,3-dipolar cycloaddition reaction.2. Synthesis of new 1,2,4-triazolo[5,4-d][1,5]benzothiazepine derivatives.A new series of 1,5-benzothiazepine derivatives containing 1,2,4-triazole were synthesized from 1,5-benzothiazepines and (phenylhydrazino) chloromethylenecarboxylates via 1,3-dipolar cycloaddition reaction.Above all, the structures of the new compounds were confirmed by elemental analysis, IR, 1H NMR and MS spectral data, and their spectral property was also discussed.
