C-PC was isolated and purified from spirulina platensis by salting out and aqueous two phase extraction. Through the optimization of the traditional salting process, identified enrichment of C-PC required for the gradient of ammonium sulfate saturation. Experimental results show that, after four-step ammonium sulfate salting out, C-PC purity was 3.55, recovery was 47.13%. To further increasing the number of salting out, the purity of C-PC was not obviously increased, but the recovery was decrease extremely. The highest purity of C-PC was 3.96, while the recovery was only 9.66% after multiple salting out.Aqueous two phase extraction is used in the purification proteins in recent years. C-PC was directly purified from crude extracts, the effects of different parameters such as type of aqueous two phase systems, phase forming salts, concentration and molecular weight of the phase forming polymer, concentration of phase forming salt, system pH, and type and concentration of neutral salts on partitioning of C-PC were studied. The type of aqueous two-phase for the separation and purification of C-PC was determined polymer/salt systems, and the phase forming salt was potassium sodium tartrate. Desirable conditions for differential partitioning were obtained in 16% (w/w) polyethylene glycol (PEG) 2000, 21% (w/w) potassium sodium tartrate, at pH 6.0. Multiple extractions resulted in further increase in purity of C-PC without loosing the yield and a maximum purity of 3.91 was achieved in third aqueous two-phase extraction. Ammonium sulfate precipitation and multiple extractions of aqueous two-phase extraction resulted in further increase in purity of C-PC, a maximum purity of 5.01 is achieved after all these separation process. The C-PC microencapsulation materials were chitosan and alginate. The optimum conditions determined by experiments were the degree of deacetylation of chitosan 74%, stirring speed 600 r/min, stirring time 30min, distance from needle end to fluid 10 cm, drop rate 30 drop/min, sodium alginate concentration 2.5% (w/v), chitosan concentration 0.6% (w/v), C-PC/sodium alginate 1/4 (w/w), CaCl2 concentration 2.0% (w/v).
