This paper mainly studied the fermentation conditions by Rhizopus chines 12th again. According to the results of single factor analysis and orthogonal experiments, the optimum medium and optimum culture conditions were determined. Inoculation were size 5×105/ml and 20% in inoculum medium and fermentation culture respectively. The optimum culture condition were flask rotation speed 120r/min during primary 24h, 150r/min during 24-26h, and 170r/min during final 24h, with pH nature, fermentation time was 60h, and the optimized medium was wheat bran abstract 5%, tryptone 1%,bran residue that the nitrogen is equal to 2% in inoculum medium and fermentation culture respectively,0.006mol/L Fe+2,0.003mol/LMn+2.Tryptone, Fe+2, Mn+2,fermentation temperature and flask rotation speed influent activity of enzyme,the activity of enzyme was increased from primary 56.26u/ml to 230u/ml.The methods of kinetics were determined primarily, that is foundation for deeply studing.
