The Isolation And Purification Of Active Components From Aucklandia Lappa Decne. And Aquilaria Sinensis (Lour.) Gilg Of Pure Detoxification Drug-Jitaipian

Narcotics abuse is a growing medical and social problem. The traditional Chinese medicine (TCM) is becoming increasingly important for the addiction treatment because of it special advantages.'Jitaipian' is a pure detoxification drug which consist of 15 traditional chinese medicines like Yanhusuo,Danshe,Chenxiang and Muxiang etc. In clinical practice, 'Jitaipian' shows significant effect for the treatment of protracted withdrawal symptoms which is very common in patients, and then gets rid of toxin and make body health.At present, the prescription compatibility, effective ingredients and active mechanism of 'Jitaipian' is not very clear. This limits the clinical application of the drug. The existing quality standard of'Jitaipian' is simple, which can not control the effective constituents of the medicine very well. Our research chooses Muxiang and Chenxiang as materials and separates active ingredients from these two medicinal herbs by phytochemistry.According to the provisions of Pharmacopoeia, we have checked on the quality of Muxiang and Chenxiang which used for research. Two known sesquiterpene lactones named costunolide and dehydrocostuslactone was separated from extract of Aucklandia lappa Decne. by silica gel. We have established an HPLC method for the determination of agarotetrol and 4'-methoxyagarotetrol in Aquilaria sinensis (Lour.) Gilg. A simple and efficient preparative HSCCC method was also established for separating agarotetrol and 4'-methoxyagarotetrol by using chloroform-methanol-water (4:2.6:2.4, v/v/v) as the two-phase solvent system. HSCCC was successfully used for the separation and purification of these two main 2-(2-Phenylethyl) chromone derivatives which have similar structure with ideal purity.
