Arsenic and mercury compounds exist widely in nature. Both of them can cause serious harm to human health. The toxicity of arsenic and mercury compounds is closely related to their forms. As for the arsenic compounds, the toxicity of inorganic arsenic is far than organic arsenic, the more arsenic combined with the organic group, the litter toxicity the arsenic has; as for the mercury compounds, the toxicity of organic mercury is far than inorganic mercury, the toxicity of alkyl mercury is far than aryl mercury. Morphological analysis is more scientific than the determination of the total content, when determined arsenic, mercury content and evaluated their toxicity. Aquatic products are the important sources which can bring arsenic and mercury compounds to human body, because they can absorb and enrich them. So it is important to establish a fast monitoring, accurate analytical method for determination of arsenic, mercury speciation. In this paper, mercury compounds in aquatic products was extracted by hydrochloric acid extraction, ultrasonic extraction method and determined by high performance liquid chromatography-atomic fluorescence spectrometry (HPLC-AFS) and high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS). The effect of hydrochloric acid concentration, high performance liquid chromatography conditions, standard solution preparation conditions and other factors were discussed, and the relative standard deviation (RSD),spike recovery and determination of reference materials were used to detect the accuracy of this method. The proposed method was used to determine the contents of mercury speciation in tuna and other seafood and aquatic products of several provinces and citys, the results of two hyphenated techniques were consistent. In this paper, the method of arsenic speciation was also proposed. Arsenic compounds in aquatic products was extracted by water-acetic acid extraction, ultrasonic extraction method and determined by HPLC-ICP-MS. Six pre-treatment methods were investigated on the effect of arsenic speciation, HPLC conditions were established, the applicability of AFS and ICP-MS were compared. The relative standard deviation (RSD) and spike recovery were used to detect the accuracy of this method. The proposed method was used to determine the contents of arsenic speciation in hairtail and other seafood and aquatic products of several provinces and citys, the results showed that this method was satisfactory.
