Using the Faint-scent type Tie kuanyin tea as experiment material, established the optimum extraction technology and purification technology of Tea polyphenols, analyzed components and content of Aromatic substances, and studied biological activity of Tea polyphenols and Aromatic substances. The main research results are as follows.1.The extraction technology was optimized by single factor and orthogonal experiments. The optimum technology of ethanol extraction of Tea polyphenols is as follows:solid-liquid ratio is 1:25, extraction temperature is 75℃, extraction time is 50min, alcohol chroma is 75%, the amount of Tea polyphenols is 267.7mg/g on this conditions.2.Tea polyphenols was purified by resin absorption. Based on comparing adsorption and desorption characteristics of 5 macroporous adsorption resins H1020, AB-8, D-101, DM301, NKA for Tea polyphenols, finded out that DM301 type macroporous resin is considered as optimum resign for purification of Tea polyphenols, established the optimum technology of purification of Tea polyphenols. Results display, when flow rate 1.5ml/min, pH value 3.0, the concentration of ethanol 80%, desorption velocity 1.5ml/min, the rate of Tea polyphenols was 15.69%, purity was 94.78%.3.The volatile oil was extracted with SDE method, and then identified and quantified by GC/MS. 56 aromatic substances were detected. Among them alcohols are 58.1%, hydrocarbons are 15.48%, esters are 9.46%, nitrogen compounds are 7.29%, unknown compounds and other compounds are 5.09%, heterocyclicoxygen compounds are 2.47%, aldehydes are 0.98%, ketones are 0.79%, acids are 0.34%, total volatile oil is 0.0073%.4.Through setting up mouse S180 tumor-bearing models, studied the anti-tumor functions of Tea polyphenols and Aromatic substances. All the dosage groups of Tea polyphenols had highly significant function on the suppression of the S180 tumor activity, negatively related with dosage, and the rates of anti-tumor were 23.81%, 29.17%, 33.33%. All the dosage groups of Volatile oil had no significant effect on the suppression of the S180 tumor activity. All the experiment groups of Tea polyphenols highly significan increased thymus index and spleen index of mouse, negatively related with dosage. Compared with control group, the thymus index and spleen index of high dosage of Volatile oil were increased by 21.11%(p<0.01), 26.03%(p<0.05).5.To calculate Half-death peroid, Mean life-span and Mean maximum life-span by Drosophila survival test, and determine SOD value, MDA content and total anti-oxidation value in 6o ageindays Drosophila.All the experiment groups of Tea polyphenols and Volatile oil could prolong the life-span of drosophila. The high dosage of Tea polyphenols could prolong the Half-death peroid of male and female by 17.18%(p<0.05), 25.07%(p<0.01), increased Mean life-span of male and female by 14.58%(p<0.05), 20.18%(p<0.01), and lengthened Mean maximum life-span of male and female by 15.11%(p<0.01), 13.20%(p<0.01). The medium of Volatile oil could significant prolong Half-death peroid of male and female, the elongation rates were 13.28%, 12.97%, significant prolong Mean life-span of male and female, the elongation rates were 12.39%, 19.32%, but had no significant effect on Mean maximum life-span of Drosophila.The high dosage of Tea polyphenols significant increased SOD value of male and female by 12.82%(p<0.01), 38.41%(p<0.05), highly significan decreased MDA value of female, but had no significant effect on male, highly significan increased total anti-oxidation value of female. The medium of Tea polyphenols significan enhanced SOD value and total anti-oxidation value of female by 9.94%(p<0.05) ,21.88%(p<0.05), highly significan decreased MDA value of female by 9.80%(p<0.01). The medium of Volatile oil significant enhanced SOD value of Drosophila, highly significan decreased MDA value of female, and increased total anti-oxidation value of female. Both the high and low dosage groups of Volatile oil had no significant effect on SOD value, MDA value and total anti-oxidation value of male and female.
