Polygonatum odoratum (Mill.) Druee is Liliaceae Polygonatum odoratum class dry roots. Polygonatum odoratum has improved blood fat, blood pressure, myocardial ischemia, alleviate atherosclerosis plaque formation, so it enable extend the biological life. In order to further develop of Polygonatum resource.this issue selected odoratum planted in Sichuan,using modern technology let Polygonatum glycoprotein isolated, purified, and purified,and has got the determination of the composition and structure of Polygonatum glycoprotein.The antioxidant activity were also studied in the test. the main research results were as follows:1. Odoratum glycoprotein was extracted by hot water,to study the four main factors. Orthogonal experiment design were as follows:the ratio of sample to water was 1:30, temperature 70℃, extraction time 4h, and the extraction times was 4, in this condition, the extraction ratio of glycoprotein was 8.28%.2. The optimum conditions with Microwave-assisted extraction were as follows: the ratio of sample to water was 1:40, extraction times of 3, microwave power was 420W, extraction time in Microwave was 3min, in this condition, Odoratum glycoprotein yield reached to 8.85%.3.Two fractions named respectively PDGP-1, PDGP-2 were isolated and purified from the Polygonatum glycoprotein by the DE-52, Purity of glycoproteins used the Sepharose CL-6B gel column chromatography, two purity fractions after gel filtration, named PDGP-1', PDGP-2', the molecular weight were 92.45kD and 60.28kD.4. The sugar content of PDGP-11, PDGP-21 were 75.22%,75.13%; protein content were:16.35%,18.42%.5. Odoratum glycoprotein monosaccharide composition analysis and IR analysis showed that:PDGP-1'in the main components of the sugar chain of glucose, galactose, with a small amount of rhamnose and an unknown component, PDGP-2' in main components of sugar chains of glucose, galactose, with minor amounts of fructose, mannose, xylose and arabinose. Two glycoproteins contain pyran ring.6. Amino acid composition analysis showed that almost two glycoproteins containing various amino acids necessary for human body. Including glycine, phenylalanine, aspartic acid, glutamic acid, proline content is high, also constitute a plant containing O-glucose glycoprotein peptide bond of serine, tyrosine. Polygonatum glycoprotein is a high quality protein.7. Antioxidant test results show that the total glycoprotein Polygonatum reducing power increased with the concentration gradually increased, reducing power and glycoprotein concentration correlation. Glycoprotein on the organic free radical (DPPH) and hydroxyl radical scavenging effect significantly, and with the increase of concentration of PDGP, scavenging ability increased.
