Synthesis, Cytotoxicity Of N-carboxy Acyl α-amino Acid Dianion And Diamine Palladium(ii)/ Platinum(ii) Mixed-ligand Complexes In Vitro

The thirty-nine palladium(Ⅱ) /platinum(Ⅱ) mixed-ligand complexes with N-carboxyacyl-α-amino acid dianion and diamine have been synthesized and characterized by elementalanalysis,IR,MS,UV and 1H NMR spectra techniques. The crystal and molecular stucture ofnine complexes were determined by X-ray diffraction. In all complexes,the palladium(Ⅱ) ioncoordinated to the N atoms of the diamine molecule, the deprotonated amide nitrogen and onecarboxylic oxygen of the amino acid molecule in a square-planar geometry as afive-membered chelate ring. The complexes have been assayed for cytotoxicity in vitroagainst cell lines:human immature granulocyte leukemia (HL-60), humanliver carcinoma(Bel-7402), human gastrocarcinoma (BGC-823) and human nasopharyngeal carcinoma (KB)by SRB(Sulfonylrhodamine B) and MTT(Methylthiazolyl tetrazolium) methods. Theresults indicated that the complexes have selectivity to cancer cells, the cytotoxicity of theplatinum(Ⅱ) complexes is not higher than the corresponding palludium(Ⅱ) complexes. Thespecies of diamine,amino acid and acyl agent have effects on the cytotoxicity.Thepalludium(Ⅱ) complexes containing 1,10- phenanthroline,N-phenylacetyl amino acid orN-phenylpropionyl amino acid have better actvity because of the different sensitivity to celllines. The anticancer mechanism need to be elucidated in molecular level.
