The analysis of the banned matters in foods is of importance for assure food safety and human healthiness. In this paper, the aim is to pure and extract samples using solid phase extraction(SPE), then apply reverse phase high performance liquid chromatography (RP-HPLC) to separate and determinate some banned substance. It includes four chapters as follows:Chapter1:The basic information of SPE was introduced briefly.Besides, a review mainly concerned with the technolgies of seperating and determinating melamine and its analogues in recent years.Chapter2:To establish a new method for effective separation and determination of melamine in liquid milk by reversed phase high performance liquid chromatography with diode array detector. The recoveries of melamine were compared on different solid phase extraction columes and in different extraction conditions.The mobile phase is confirmed by regulation of the component of buffer solution and the proportion of organic solvent and water, and melamine in samples were separated excellently. The method was convenient, rapid and accurate.Chapter3:To establish a method for effective separation and determination of cyromazine and its metabolite melamine in liquid milk and eggs by reversed phase high performance liquid chromatography with diode array detector. The retention behavers of different mobile phases were discussed in order to choose appropriate ion pair regent;The conditions of sample pretretment and liquid phase seperation were optimazied to decrease LOD and LOQ, which could achieve effective separation and determination of cyromazine and its metabolite melamine.Chapter4:To establish a new method for effective separation and determination of malachite green,enrofloxacin and ciprofloxacin in aquaculture water bodies and fish feed by reversed phase high performance liquid chromatography with solid-phase extraction. The recoveries of three analytes were compared on different solid phase extraction columes and in different extraction conditions. The mobile phase is confirmed by regulation of the component of buffer solution and the proportion of organic solvent and water in order to separate three analytes in real samples excellently. The method was novel, accurate and reliable.
