In this paper a total of 156 strains were screened from the soil, 38 strains from LB medium, 51 strains from PDA medium, 67 strains from Gao at medium, there were 33 strains to be able to grow on chitin medium,Screen from the soil were four high-yielding strains of chitinase, Enzyme production capacity from high to low strain number followed by C65-2, L63-4, G15-2, G38-4, on the four bacteria strains were identified, Results as follows: C65-2 for Aspergillus fumigatus, L63-4 for Paenibacillus sp, G15-2 for Pseudomonas aeruginosa, G38-4 as an Aspergillus, the chitinase producing conditions was optimized for Aspergillus fumigatus C65-2, the results of optimizing seed was that more use of the carbon source is glucose, more use of the nitrogen source was corn syrup, The optimal initial pH 4.96, The highest cell dry weight after 36h, he results of Optimizing Enzyme production medium was that Enzyme inducer activity for the chitosan was the highest, after 42h fermentation activity of the highest, Initial pH 6.0 the highest activity, Experimental results show that Chitosan reached 2%, the phenomenon disappeared cabbage, the maximum activity could be up to 6.9 U/ml and the enzyme activity increased 210% than before. The study of enzymatic properties of the chitinase showed that the molecular weight was 20 Kda. Chitinase maximum absorption wavelength of 207, thermostable stability of the chitinase was not good and the enzyme activity reduced to 0 at 60℃for 50 min. The optimal enzyme reaction temperature was 55℃and the optimal pH of enzyme reaction was 7.0. The chitinase activity can be enhanced by Mg2+, Cu2+ and inhibited by Fe3+, Enzyme product was more than two sugar chitooligosaccharides.L63-4 is a Bacillus, it was more conducive to preservation, easily degraded, its hydrolysis products were also rich, therefore, L63-4 was a more promising bacteria. G15-2 strains were special forms, start color was white, then gradually turn red, and finally into ink blue, Even and fully dyed blue agar, after the literature search, microorganism which could secrete a dark blue pigment was very rare, therefore, G15-2 could serve as a pigment producing strains of microorganisms.
