Microecosystem of pit mud was an organic whole which depended on the interdependence and interaction of microbial community and ecological factors.The maturation process of mud pit was the process of gradual evolution of micro ecosystem.The degree of maturation process determined the quality and style of Luzhou-flavor Liquor.Previous researches of mud pits unilaterally focused on,such as analysis the component of mineral elements, analysis of physical-chemical compositions, cultivation and identification of microbial strains,and so on. Few studies had involved the entire microflora evolution of mud pit. In this study, different aged pit muds from LuZhouLaojiao were sampled to study the evolution trend of mineral elements, microbial metabolism and microbial community diversity with the maturation process of pit mud by using X-Ray Fluorescence(XRF), Gas chromatography-mass spectrometry(GC-MS) and Single-strand conformation polymorphism(SSCP) analysis technology. The results indicated that:1. Ecological factors was showed regular change : (1) The moisture contents of different aged Pit muds showed a rising trend with increasing of pit age;(2) The pH value of all those Pit muds were weak acid,and curve of ammonia nitrogen content was the same as pH value;(3) The ammonia nitrogen content of different aged Pit muds kept about 3000mg/Kg in different pit age;(4) The humic acid content of different aged pit muds showed a reducing trend with the increasing of pit age;(5) The total nitrogen content of different aged pit muds showed a reducing trend with the increasing of pit age;(6) The Ca,K,Zn content of different aged Pit muds showed a rising trend with increasing of pit age;(7) The Fe,Mg,Mn,Al content of different aged pit muds showed a reducing trend with the increasing of pit age;(8) The Ba,Cl,Cr,Rb content of different aged pit muds showed a first reducing then rising trend with the increasing of pit age,and were lowest in 100 pit ages;(9) The Cu content of different aged pit muds kept about 40mg/Kg.S,P contents of those pit muds over 100 pit ages were higher than that of 20 pit ages.2. Regular change were showed in metabolic products of microorganism of pit mud:(1) The alcohols content of pit mud showed a reducing trend with the increasing of pit age,and slightly toxic substance–diethylene glycol was found in young aged pit muds;(2) The variety and content of ester in different aged pit muds showed a rising trend with increasing of pit age;(3) The aldehydes content of different aged pit muds showed a reduing trend with the increasing of pit age;(4) Only palmitic acid was found in upper pit mud of 100 pit ages.3.Community structure of eubacteria in pit muds:(1) Results of abundance of SSCP spectrogram showed a larger change in community structure of eubacteria in different aged pit muds with the increasing of pit age;(2) Results of dominance of band of SSCP spectrogram showed a larger change in dominance of band,and ecological relationship of mutualism or competition between some bands;(3) Results of eubacteria Shannon index of SSCP spectrogram showed a rising trend with increasing of pit age,and showed stability and resistance of microecosystem of pit mud improve;(4) Results of similarity of SSCP spectrogram showed a larger change in community structure of eubacteria in pit mud with the increasing of pit age.4.Community structure of archaea in pit muds:(1) Results of abundance of SSCP spectrogram showed a larger change in community structure of archaea in pit mud with the increasing of pit age;(2) Results of dominance of band of SSCP spectrogram showed a larger change in dominance of band,and ecological relationship of competition between 2 bands of archaea.Compared dominance of archaea bands with eubacteria', and found No.19 band of archaea and No.9 and 19 band of eubacteria showed ecological relationship of Mutualism;(3) Results of archaea shannon index of SSCP spectrogram showed a rising trend with increasing of pit age,and showed stability and resistance of microecosystem of pit mud improve;(4) Results of similarity of SSCP spectrogram showed a larger change in community structure of archaea in pit mud with the increasing of pit age.
