Protein is the substances,which widely exists in the body. The most abundant protein is serum albumin in the blood plasma. The studing on the interaction between serum proteins and small molecules have significant meaning for the life sciences, pharmacokinetics and the mechanism of drug action.The thesis investigated the interactions of some compounds with serum proteins by using fluorescence and ultraviolet spectroscopy, overlap integral method and the reaction thermodynamics. The results can give the information about the mechanism of the interactions between biological macromolecules and drug, pharmacokinetics and reaction thermodynamics. The main contents are as follows:The first chapter reviews the applications of spectrocopy on the interaction of biological macromolecules with small molecules.Chapter II, the interaction between benzbromarone and Bovine Serum Albumin was studied by spectroscopic methods, under simulative physiological conditions. The quenching mechanism and their binding sites were deduced according to Lineweave-Bure and the Stern-Volmer equation. The type of acting force was obtained, based on the thermodynamic parameters.Chapter III study Buflomedil hydrochloride with human serum albumin interaction, the results show that the quenching mechanism may be a static quenching mode. The electrostatic interaction was the mainly binding force. The effects of some metal ions on the binding were studied.The fourth chapter discusses the interaction of the pyran derivatives4-nitrophenyl-(di-4-hydroxy-6 -methyl-2-oxo- 2H-pyran-3-yl)methane (NPPM)and 4-fluorophenyl-(di-4-hydroxy-6-methyl-2-oxo-2H- pyran-3-yl) methane (FPPM) with human serum albumin (HSA).The mechanism might be a static quenching.Van der Waals force and electrostatic binding might play main roles for NPPM-HSA and FPPM -HSA, respectively. The effects of some metal ions on the interactions between the small molecule and HSA were discussed.Chapterâ…´,studing the interaction of nucleoside compounds,5-(ethyl2-amino-4-methyl-5-oxo-7- phenyl-5,6,7,8-tetrahydro-4H-chromene-3-carboxylate)-2'-deoxyuridine(EAMD)and 3'5'-di-0-acetyl-5- (2-amino-7,7-dimethyl-5-oxo-5,6,7,8-tetrahydro-4H-chromene-3-carbonitrile)-2'-deoxyuridine(DAAD) which possess biological activities with protein. Its quenching mechanism may be a dynamic quenching. The acting force is mainly hydrophobic interaction force.
