Lysome, as a kind of antibacterial protein that acts on the cell wall of microorganism, is used in the feed, food and medicine industry and so on extensively because of its strong antibiotic action. But, the problem is raised that the lysozyme can not resistant to the acid in stomach when it is used in the gastro intestinal digestive system. To the problem, the paper studied the preparation of lysozyme microcapsule by the internal emulsification method. The chitosan-alginate was chose as the microcapsule wall. Also the control-release property of the microcapsule of chitosan-alginate sodium in imitated stomach-liquid environment and the activity of embedded lysozyme in the imitated stomach-liquid were determined. The paper studied the effect of microcapsule prepared by colza oil instead of paraffin oil to make for making food grade microcapsule. At last,in order to make the feasibility of the technology better,the paper firstly studied on recycling the oil including colza oil and paraffin oil . The key results are as follows:When the concentration of alginate sodium, chitosan and lysozyme were 2%, 0.3%, 1.5% respectively, and the volume ratio of Span80 to oil was 1:100, the result from orthogonal experiments was that the mass ratio of CaCO3 to alginate sodium was 7:40, and the ratio of water to oil was 30:70, aand the mass ratio of glacial acetic acid to CaCO3 was 3:1.The results of the property of microcapsule prepared under the best technique are: the entrapment efficiency was 89.2%, the loading efficiency was 34.9%, the release ratio in imitated stomach-liquid after 2h was 35.5%, and the whole release ratio in imitated liquid after 4h was 88.9%, the activity retention rate of embedded lysozyme in the imitated stomach-liquid was 89.5%, but that of lysozyme which was not embedded was 31.7%. The activity retention rate of embedded lysozyme was improved significantly.The property of lysozyme microcapsule prepared by paraffin oil and plant oil respectively were: the entrapment efficiency were 89.2% and 81.7%, the loading efficiency were 34.9% and 26.6%, the release ratio in imitated stomach-liquid after 2h were 35.5% and 38.7%, the whole release ratio in imitated liquid after 4h were 88.9% and 85.4%, and the activity retention rate of embedded lysozyme in the imitated stomach-liquid were 89.5% and 84.7%. The effect of the technique prepared with plant oil was lower than paraffin oil.The methods of recycling of paraffin oil and canola oil were studied. To recycle the paraffin oil, through the compare of heating method, the heating method with the add of proper volume of mineral salt, and the heating method after centrifugation, at last the third method took the shortest time on recycling the paraffin oil. The centrifugation condition was that the rotate speed was 10000rpm, the time was 10min, and then heated in 60℃water bath 5min. Compared to paraffin oil, the plant oil could be recycled more easily. The condition was that the rotate speed was 10000rpm, and the time was 10min. At the end of the paper, the effect of recycled paraffin oil and canola oil were determined. The results were: the entrapment efficiency were 86.7% and 80.5%, the loading efficiency were 33.8% and 25.3%, the release ratio in imitated stomach-liquid after 2h were 37.1% and 39.5%, the whole release ratio in imitated liquid after 4h were 85.9% and 84.6%, and the activity retention rate of embedded lysozyme in the imitated stomach-liquid were 87.4% and 83.8%. The effect of the technique prepared with recycled oil was a little lower than the primary oil.
