As a new near-infrared fluorescent dyes, aza-BODIPY dyes have attracted great attention in the last ten years. Aza-BODIPY bearing excellent emission and absorption wavelength, good photostability, sharp and intense absorption, high fluorescent quantum yields and great molar extinction coefficient, was extensively applied in biological analysis.This paper mainly elaborates the modification by introducing hydroxyl, carboxyl and alkynl groups in the large conjugated aza-BODIPY core. Besides, a novel aza-BODIPY structure with styryl group was- synthesized for the first time. Moreover, 1H-NMR, 13C-NMR, IR, MS were employed to identity the structures of the aza-BODIPY dyes. Spectral properties of these dyes were well studied afterwards.In recent years, aza-BODIPY dervatives with special photophysical and photochemical properties had been widespeadly used in fluorescent sensors, biolables. Howerver, the prerequisite for the application is how to synthesize aza-BODIPY with special functional groups, such as carboxyl and hydroxyl. In chapter 2, we have synthesized aza-BODIPY dyes with hydroxyl, carboxyl, and alkyne and so forth. The demethoxylation of 6-methoxy-3,4-dihydronaphthalen-1(2H)-one generated the hydroxyl groups, followed by respectively protecting by TBSCl and t-butyl bromoacetate. The corresponding aza-BODIPYs were synthesized. Furthermore, the modifition was carried by the deprotection of aza-BODIPYs to generate hydroxyl or carboxyl functional groups.In order to make the fluorencent of the dye bathochromic shift, a direct way is to extend the conjugation of the aza-BODIPY core. In chapter 3, 4-free pyrrole and five kinds of pyrroles with sytryl system were synthesized. what's more, the confocal fluorecent image of U87 glimoa cells treated with BODIPY 32 demonstrated good result and potential utility.
