1,3,4-thiadiazoles derivatives are widely investigated and applied to medicine, agriculture, industry and so on, especially on the progress of synthesize anticancer medicine. In this paper,5-amino-1,3,4-thiadiazole-2-thiol was used as starting material to modify the 2-metcapto and 5-amino so as to obtain new compounds and their corresponding synthetical method. The new compounds were expected to provide the basis for drug synthesis and screening. The main content was described from three aspects as follows:1. Optimum condition for the AMT's Michael addition reactionThe Michael reaction can react at thiol group and 3-N of AMT, and two types of product obtained. Take the reaction of acrylonitrile as a example, the author studyed substrate mole ratio, reaction time, reaction temperature, reaction solvent with it's dosage, catalysts with it's dosage and take use of anti-oxidation measures respectively, and then got the favorable bonus conditions for each product. Finally, the yield of a1 reach to 46.74%, and 49.37%for b1.2. The applicability of AMT's Michael addition reactionTake AMT as starting material, the author changed several electrophile so as to react Michael addition, and sum up serviceable range of this reaction. Besides, six new thiadiazol derivates which haven't been reported were obtained, and each of them was confirmed by IR and1H NMR spectra.3. Imidazolidinyl and thiazolidinyl thiadiazol derivates were synthesized successfullyThere is a naked aminal-group in each compound al-a3,b1-b3, which got from AMT's Michael addition reaction. Accourding to this character, these compounds after acylated with chloroacetyl chloride and cyclizated with potassium sulfocyanate in the condition of catalyzator, six imidazolidinyl thizdiazol compounds and six thiazolidinyl thiadiazol compounds were obtained and each of them was confirmed by IR and1H NMR spectra.
