This thesis mainly includes three parts:The first part summarized the synthesis and application of chiral spirocyclic ligands in asymmetric catalysis over the past decade years; The second part described the attempts to synthesize the chiral spirocycles via intramolecular alkylation under the asymmetric phase transfer catalysis and via intramolecular Stetter reaction of nitroalkenes under N-heterocylic carbenes; The third part was about the synthesis of functionalized indole intermediate, one key building block for the synthesis of Aza-galanthamie, wherein the main synthetic steps included the phenolic oxidative coupling, the alcoholysis as well as the aza-Michael addition.
