Organic fluorescence probe is one of the important fine chemicals, it has its unique properties, and play very important roles in the fields of metallurgy, agriculture, archaeology, mineralogy, environmental chemistry, pharmacology, physiology, biology, molecular genetics, information science, fluorescent marker and laser dye. In recent years, with the continuous development and perfection of the organic fluorescence probe, this will bring new development opportunities for genomics, proteomics, biological chip and drug mechanism, and it also provides valuable methods and information. With the improvement of the synthesis and technology, people's awareness of organic fluorescence compounds will further. The scope of its application will expand and the development of fluorescent probes will be more extensive, so looking for more effective organic fluorescence probe becomes one of the most popular researches. In our research, we designed and synthesized a series of pyrido[3,2-a]phenazine derivatives, and performed the preliminary fluorescence activity, antitumor activity and DNA Top1 inhibited activity research.This dissertation includes three parts:1. A series of pyrido[3,2-a]phenazine derivatives were designed and synthesized. Furthermore, we improved the efficiency of the intermediate (DCQ) synthesis, through the improvement, in not reduce production rate,we shortened the reaction time.2. We studyed the fluorescence test of pyrido[3,2-a]phenazine derivatives, we fine that these compounds have good fluorescence activity,which it accord with design expected. And the F value decreased in with the increasing of solution, it is different from any conventional fluorescence compounds.3. We evaluated for their cytotoxic activities toward Hela cells, lung adenocarcinoma cancer (GLC) cells, leukemia (K562) cells by MTT method. Most of the compounds show significant cytotoxic potency; at the same time, we also finded these compounds have good indirect inhibited activity to the DNA topological isomerase I.To sum up, this type of small molecules have good fluorescence properties,and have a potential application field in cancer treatment.
