Objective:Strychnine is an alkaloid from seeds of Strychnos. Strychnine is the main active component in Strychnos, is also toxic components. The therapeutic dose and toxic dose is very close, limiting its clinical application. At present, strychnine nitrate formulations are injections of strychnine and strychnine tablets. The safety range is small, and easy to convulsions, and is accumulative. To study on the strychnine of transdermal drug delivery system to explore the feasibility of its preparation Cataplasmata, to provide a new route of administration.Methods and Results:1. Study on transdermal delivery of StrychnineThe methods of High Performance Liquid Chromatography (HPLC) was established determined the strychnine in the solubility in different solvents. On the basis of preformunlation, the transdermal delivery of strychnine across excised pig skin was studied with Franz diffusion cell. The results showed that there was no significant difference of strychnine in vitro permeation of ethanol effect. The effects of different penetration enhancers on strychnine percutaneous penetration of in vitro results showed that the terpinolene and limonene has a strong penetration effect. The strongest of penetration is terpinolene.2. Study on the preparation for strychnine cataplasmataThe formulation of strychnine cataplasmata was carried out and optimized by orthogonal design method. The strychnine cataplasmata was evaluated by adhesive force and holding force, the optimun proportion is:NP-700 1.7g, dihydroxyaluminum 0.3g, citric acid 0.3g, glycerol 15g. The prepared according to the optimized formulation has a good poultice physical property. Terpinolene used as penetration enhancers, study on the transdermal delivery of strychnine cataplasmata. established a rapid and sensitive High Performance Liquid Chromatography (HPLC) assay to measure the content strychnine in the strychnine cataplasmata, the release and permeation in vitro. The release pattern of strychnine cataplasm followed weilbull distribution.
