Extraction and modification of wheat bran dietary fiber were mainly studied in this paper. The three drying methods were compared screening to receive the best way to dry.The contrast experiments were design to get the method of extraction of wheat bran dietary ,and optimized the process parameters. Based on this research ,we studied the effect of dietary fiber quality of the colloid mill superfine grinding modification and enzymatic modified.The experiment researched the influence of physical properties of dietary by seven drying ways according to the bran state, insoluble dietary fiber extraction,WHC and SWC. Results showed that microwave drying which needed the shortest time was superior to the other indexes of drying methods. The WHC and SWC of microwave drying extraction prandial fiber were achieved 6.5 g/g and 5.8 mL/g. In addition, microwave drying of water-soluble dietary fiber from the extraction of have obvious auxiliary function.Experimental results showed that two-enzymes method extracting dietary fiber was 0.64% more than enzyme-chemical method in protein residues.So using enzyme-chemical method to deal with protein was more thoroughly. But enzymatic was 10% higher in IDF extraction and respectively higher 26.3% and 8.7% than enzyme - chemical method in WHC and SWC.The paper determined the wheat gluten extraction for 40 mesh size acccording to the impacts of IDF extraction yield ,WHC and SWC. By single factor and orthogonal test, the high-temperature resistant amylase enzymes hydrolyzation on starch can draw the optimal preparation conditions:high temperature resistant amylase enzymes additiion0.5%, under the condition of pH6.5,temperature84℃,hydrolyzing time 45min and extraction rate of IDF is 92.8%.Alkali protease zymolysing protein optimal technological conditions through orthogonal test comprehensive balance method is protease enzyme addition1.0mL(120u/g), hydrolyzing time 100min, temperature 64, pH8.0,and IDF extraction yield reached 94% ,WHC was 6.80g/g and SWCwas 6.0 mL/g.The optimal conditions of hydrolyzation on dietary fiber with wet superfine grinding modification were got from single factor and orthogonal test.While colloid mill repeated grinding number five times, solids content for 80%, gear clearance fixed in 25 mm, the results were WHC 10.8 g/g, SWC 6.3 mL/g,oil absorption force for 3.1 mg/g,viscosity 20mPa·S or so,and cholesterol absorption force in pH = 2 and pH = 7 respectively were 2.9 mg/g and 4.3 mg/g. The content of SDF was about 16%.The experiment adopted compounded cellulase enzymes and xylanolytic to modify dietary fiber ,and the highest rated optimal conditions were analyzed from comprehensive assessment of experimental results .In the condition of cellulase addition for 0.05 mL (40U/g), xylanase adding amount of 0.06 mL (48U/g), material to liquid ratio 1:10, enzymatic hydrolysis time 5h, the results were that quality of SDF was 3.5729 and ratio for 35.73%; WHC of DF was 12.6 g/g, SWC was7.9 mL/g, oil absorption force was 4.2 mg/g.The physical properties of dietary fiber modified by enzymatic combination of wet superfine grinding wereWHC13.9g/g,SWC8.2mL/g,oil absorption force 4.6 mg/g, respectively improved 2.04times and 1.37times, and viscosity was 12.3 times than the unmodified averaged .
