Numerous studies show that probiotics can adjust the intestinal flora,enhance immunity,and lipid-lowering function.The purpose of this research is the further investigation of the compound lactobacillus microcapsules performance and regulation of the intestinal flora in rat with high-fat diet.The low methoxyl pectin(LMP) was used as wall material,Lactobacillus fermentum and Lactobacillus plantum in combination were encapsulated into the microcapsules by ionic-crosslinking process.After freeze drying, the encapsulating rate was more than 99% and survival rate after lyophilization was 85%. The in vitro tests in simulated gastrointestinal fluid showed that the gastric ?uid and bile juice had a destructive effect on the naked Lactobacillus plantarum or fermentum bacteria and mixed naked bacteria, with 4.8,5.0 and 4.94 log cfu decrease. While the number of microcapsule loading Lactobacillus plantum or Lactobacillus fermentum or both bacteria decreased by 1.66,1.75 and 1.78 log cfu,respectively.The storage stability test showed that the best storage temperature of the microcapsules as-20 ℃,indicating the protective effect of microcapsules on Lactobacillus fermentum or plantum.The results from effects of compound Lactobacillus microcapsules on lipid-lowering effect in rats fed with high-fat diet showed that at the end of 6 week, the number of probiotic flora such as Lactobacillus and bifidobacterium in high-fat diet group is significantly lower than that of the control group(P<0.05), the group of microcapsules is higher than the naked bacteria groups(P>0.05).the number of enterobacter in fat control group is significantly higher than others(P>0.01).and enterococcus is no significant difference in each group(P>0.05).Contents of serum TG and TC were significantly higher in fat control group than the control group(P>0.05).IL-6 is significantly higher than the control group(P>0.01),the group of microcapsules is lower than the naked bacteria groups(P>0.05),SIg A is significantly lower than the control group(P>0.01),the group of microcapsules is higher than the naked bacteriagroups(P>0.05). ET is significantly higher than the control group(P>0.01),the group of microcapsules is lower than the naked bacteria groups(P>0.05).HDL and TNF-a are no significantly difference in each group(P>0.05).Immune organ index is lower than those of high-fat groups in the control group(P>0.05).the number of Lactobacillus fermentum,Lactobacillus plantarum in rat intestinal tract has been proliferated with RT-PCR.The results showed that: compound Lactobacillus microcapsules can regulate intestinal flora and systimetic immune function in rats with high fat diet.It laid the foundation for the further development and application of composite Lactobacillus microcapsules.
