Angelica keiskei is a kind of green plant which can promote the metabolism of physical, enhance immunity, comprehensive nutrition and longevity. Angelica keiskei contains a very precious unique substance called “Chalcone”. Chalcone is a kind of flavonoid compound, which the chemical structure can be named of 1,3-diphenyl-propenone.In this article Angelica keiskei was used as raw material the green process was proposed: Angelica keiskei → extraction by Alcohol-water → chlorophyll abstraction by Hexane→tannin removed by lime→Chalcone was extracted by acetate→ Chalcone was absorbed by AB-8 Macroporous resin→ carbohydrate eluted by water→ Chalcone gradient eluted by Alcohol-water→refined Chalcone. Single factor experiment was used in the paper, ratio of material to solvent、ratio of extracting solvent、extracting time、extracting temperature and so forth factors were considered to determine the optimum process. Microscope was applied for watching morphology, ultraviolet,infrared, high pressure liquid spectrometry, and mass spectrometry was used to determine their structure. Final results are as follows: solvent ratio of ethanol: water=19:2, at71 ℃, refluxing extraction 9 h,silica gel column chromatography, n-hexane: ethyl acetate = 3:1 to recrystallize and the purity of light yellow crystals reach 6.5% was obtained.Secondly, in order to obtain higher purity of Chalcone compound the above green manufacturing technique was optimized. These factors of Ultrasound-assisted, Soxhlet extraction and the polarity of Macroporous resin were discussed to explore the effect for extraction and separation. The results show that: the purity of crystal up to 90% under the conditions that microwave-assisted soxhlet extraction with medium polarity of macroporous resin.Finally, the Chalcone content of different parts from Angelica keiskei were discussed according to the optimum green technology process and antibacterial activity was determined. The consequence show that: the highest content of Chalcone from Angelica keiskei is stem juice and the extract has antibacterial activity.
