The content of microorganisms are rich in the deep sea, and the study of marine microorganisms is of great significance in the research of biology, chemistry, material medicine, ecology, and so on. The microbial biomass in the deep sea can be detected through the testing of ATP, and this method plays a great role in the measurement of marine microorganisms. So, the design of an in-situ on-line analytical apparatus underwater is indispensable to detect the marine microbial biomass accurately, real-time.This paper summarizes the various research techniques and results in biological area of the whole world. Basing on the ATP bioluminescence reaction detection principle, with the comprehensive use of advanced technology, such as the biology, biochemistry, optics, machines, electronics, computers and so on, we accomplish an in-situ process analyzer instrument for organism detection underwater, which integrates ATP bioluminescence reaction detection method, electronic detection and computer technology.Some tests and experiments are performed, and the results show that the system noise is less than 0.008mV, the detection precision can reach 10-11mol/L. Besides, the detected error is about±10%when measuring standard solution, and the stepper precision is less than 3.6°. All of these standards have been in the leading level of the world.The dissertation is divided into six chapters:The chapter 1 expounds the background and signification of the study, and also the current developments in correlative fields about the microorganisms. This chapter stresses the characteristics of current methods and techniques, highlights the detection principle of ATP bioluminescence.The chapter 2 expounds the outline of the in situ process analyzer instrument for organism measurement underwater basing on the ATP bioluminescence reaction detection principle. And the microprocessors used in the system are also discussed. The chapter 3 expounds the most important departs of the instrument and makes a detailed introduction of the selection of the main apparatus.The chapter 4 expounds the main software of the instruments.The chapter 5 expounds the experiment results of the instrument.The chapter 6 expounds the review and future outlook of the technique.
