Due to the implementation of"confocal"technology in confocal microscopes, they perform better definition and signal-to-noise ratio (SNR) than ordinary optical microscopes, and operate 3D imaging for the samples. Laser confocal scanning microscopes (LCSM) have been widely applied in the fields of biomedicicine, material science, microelectronic manufacture and precise measurement.Nowadays, most of the confocal microscopes adopt dot confocal scanning. In 2005, the theory of line structure light confocal microscopy was put forward firstly in China by the Institute of Opt-mechatronics of Guilin University of Electronic Technology and verified by experimental research. Though the lateral resolution of line structure light confocal scanning microscopy can only reach or approach the level of the traditional dot scanning confocal microscopy, it presents two advantages compared with traditional dot confocal microscopy. Firstly, by substituting line scanning for dot scanning, the plane imaging only needs one-dimensional scanning which not only improves the velocity of imaging, but also simplifies the scanning mechanism. Secondly, the transfer quantity of light field energy is greatly enhanced by substituting detection hairline for detection pinhole, which leads to the application of the low illumination CCD for collecting images instead of the expensive photoelectric intensifier.Based on the theory and experimental research on the line structure light confocal microscopy, this thesis further studies the its key techniques and implements them into the instrument which gives rise to the development of an experimental prototype. The concrete research works of this thesis are listed as follows:1. The theory of line structure confocal scanning microscopy is further studied and its key techniques of imaging are presented accordingly.2. The special optical system for line structure light confocal scanning microscopy is designed based on the ordinary microscopic imaging optical system.3. The working plane station of high definition mechanical moving scanning controlled by open-loop and stepping motor control system are developed substantially.4. The scanning fusion software for line structure light confocal microscopy is programmed using Visual C++ programming plane. The experiment of line structure light confocal scanning microscope imaging demonstrates that the experimental prototype has a better imaging quality than the traditional optical microscopes, which has verified the theory of line structure confocal microscopy and establishes firm foundation for the development of commercial prototype in the future.
