Manganese was removed through the bio-oxidation of manganese-oxidizing bacteria in Mn biological-removal. Therefore, manganese-oxidizing bacteria in the bio-filter played a central role in the process of manganese biological removal. The contents about this research had included: isolation and identification of the bacteria in the manganese removal bio-filter, qualitative and quantitative detection screening the manganese bio-oxidation predominant strains; choosing one strain which has the most strong bio-oxidation ability to research factors, separation andpurification of manganese oxidase. 284 strains of bacteria which had been separated and purified from manganese removal bio-filter belong to 26 genus, 53 species, and Arthrobacter species is the most.Using the LBB indicator method, bio-oxidation ability of manganese bacteria which were screened from 59 species of bacteria belong to 6 genus, 14 species through qualitative analysis; 5 strains bacteria with strong oxidation ability were obtained in 10 strains of bio-oxidation ability of manganese bacteria through quantitative analysis. Arthrobacter Mn-Fe-118 was selected as the object of study follow-up test.Studied on the growth properties of Mn-Fe-118 and factors of manganese oxidase, the results showed that: as time increases, OD600 values of bacteria decreased after increasing, while the pH value has been stable growth all the way; Iron, manganese, copper, calcium four metals in a certain concentrations range could promote the activity of manganese oxide, but the activity of manganese oxide will decrease or even inactivate outside this range; iron is the most important factor affecting the activity of manganese oxide, followed by manganese, copper ions and calcium ions; optimal combination of four metal ion concentration was: Ca2+ 1.0mmol/L, Cu2+ 20μmol/L, Fe2+ 0.5 mmol/L, Mn2+ 10mg/L.The holoenzyme of Mn-Fe-118 obtained by sonication was roughly segregated by ammonium sulfate salting-out method, obtaining salting range: saturation of ammonium sulfate 40%-60%; enzyme obtained by salting-out method is separated into three clear bands, which is probably the purpose of protein with manganese-oxide activity.
