On the biomechanics experiment of loading different mechanical stimuli to culturing cells in vitro, the cell's growth condition and the change in morphology, in different time and under different load distribution of the dynamic or static stress, need to be observed and recorded. Because of the long course of the cell's cultivation and the large field of vision of the observed object, if the traditional optic microscope would be used, the experiment would be depend on operator deeply, and the operation would be also fussy. It can't meet with the demand of perennial observation for different position and different flow field, continuous tail and accurate making position. The purpose of this paper is to develop the platform of traditional optic microscope so as to implement the auto-control of the 3-D movement for microscope's carrier, auto focus, position recurrence, and this system can also repeat observation and make record for multiple objects in the large vision field.To implement the auto control of the microscope's carrier, the step motors were installed on the inverted optic microscope. The contact of flexible screw was designed to make sure the calm movement of the carrier. The single chip system controlled the step motor and host computer communicated with the single chip system by RS232 serial port, so the microscope carrier can move according to the customer's setting.The microscope control software was developed using Delphi 6.0. It can actualize multiple functions such as RS232 serial communication, image real-time display, image acquisition, autofocus and position recurrence. The method of image acquisition based on the VFW has no special definition of the image acquisition facility and the hardware of computer, so it has general application.The sharpness evaluation function of auto focus and search tactic was discussed based on the principle how blurred by the lens system in the chapter of the auto focus algorithms. To contrast the capability of these focus evaluation function, a series of tests were taken on the algorithms of auto-focus by different objects with surface roughness and variety of texture.Finally, this paper discussed the key technology of the image mosaic, and introduced the image mosaic algorithm based on the template matching, then the improvement of adaptive selecting of standard image and the muti-resolution technique was presented, and the method of seamless image mosaic was also introduced.This system offered a convenient and creditable method of image observation for the loading different mechanical stimuli to culturing cells in vitro. It can meet with the demand for tailing and recording the enormous cells' growth conditions.
