In recent year, optical imaging have become the basis for a high level of research activity toward the development of novel, noninvasive technologies for tissue diagnostics, which frequently called "optical biopsy". In contrast to histopathological examinations, optical biopsy is non-invasive and it do not require sampling by a thin-needle biopsy and material amount is not limited, radiation is delivered and received by fibers and signals are measured in a real time, the same areas can be also analyzed repeatedly.In the dissertation, based on researching medical optical biopsy of early nasopharyngeal carcinoma, we design a novel fluorescence imaging system to diagnosis and localization of nasopharyngeal carcinoma.In chapter one, the significance of diagnosis and localization for early NPC(Nasopharyngeal Carcinoma) was discussed for successful treatment of NPC. Next the developmental current status and perspectives have been reviewed in the world spectrum.In chapter two, a novel optical biopsy system based on laser-induced fluorescence for the diagnosis and localization of early NPC has successful developed, which coupled the excitation light into the original white-light transmission system of commercial nasal endoscope and characterized by coaxial imaging for both the fluorescence image and twilight image from the same targets.In chapter three, the color information of NPC fluorescence image was eliminated in that the fluorescence image obtained through the image intensifier is monochrome.In chapter four, we demonstrated the satisfied effectiveness and performance of this system by simulated NPC optical phantom, BALB/c-nu Nude Mice. In the end, we used the system in clinical test.In chapter five, the important and innovative points of the dissertation are summarized.
