Because of the peculiarity of visualization, expeditiousness and facility, Fluorescence Microscopy Technology has become a cogent method in the Life Science Research .However, the technical research and manufacture of monochromator are lagged in our country. This thesis described simulating and analyzing of new monochromator by the mathematical model, and that new demo device has been implemented.In the monochromator, short-arc Xenon lamp is employed as the light source, the all light is converged at a spot as a point light source by the elliptical mirror. During the course of collimation, grating diffraction, focus and coupling, the monochromatic light output from the optical fiber. The light power and spectral purity are the tow technical essentials of the monochromator. By the simulation of mathematical model of new monochromator system, the optical and mechanical design could be made. Whole system was developed and tested and eligible results were received.The research content in this thesis could be divided into three parts:In the first part, the principle and structure of monochromator, and the method of choosing and design of the optical components were introduced.In the second part, the energy efficiency was analysed and calculated using the optimization algorithm and optical simulation, and the elliptical reflector for monochromator was designed.In the last part, the principle and methods of the mechanical design and the process of the installation and debugging were described. The practicability is represented in the tests.
