| Triticum aestivum is the second most important crop in China and its production and quality are directly linked with both international and domestic affairs. The mainly study nowadays is a research focus in enhancing the production and quality of wheat by exogenous substance. Effects of CRZ-1 on the growth of wheat seedling under different condition were investigated. We came to the conclusion that CRZ-1 contributed to the growth of wheat seedling and the effect of 0.1 mg·L-1 CRZ-1 was the most significant. The main research methods are as follows:Firstly, the research aimed to develop an effective method to load NO fluorescent probe into plant cells. In the same concentration of fluorescent probe, fluorescent probe was loaded for different time and different temperature into plant cells, and the different fluorescence intensity was observed by fluorescence microscopy. As the loaded time increased and the loaded temperature decreased, fluorescence intensity was enhanced. It was conducive to load NO fluorescent probe with the increase of the loaded time and the decrease of the loaded temperature.Secondly, effects of CRZ-1 on the growth of wheat roots were investigated in this study. The production of nitric oxide in wheat root was detected using DAF-FM DA fluorescent emission. The intensity of fluorescent in the presence of 0.1 mg·L-1 CRZ-1 was the most increased. IAA oxidase activity decreased with all treatments of CRZ-1 from 0.01 to 10 mg·L-1. At the relatively lower CRZ-1 concentration of 0.1 mg·L-1, the activity of IAA oxidase displayed a remarkable decrease, being 40.1% lower than the control. Meanwhile, CRZ-1 treatment could increase the activities of reactive oxygen scavenging enzymes, such as superoxide dismutase and peroxidase, as determined using non-denaturing polyacrylamide gel electrophoresis. These results indicated that CRZ-1 contributed to the growth of wheat root.Finally, effects of CRZ-1 with different concentrations on the growth of wheat leaves under 20% PEG-6000 drought stress were investigated. The results showed that, the length of primary root was increased by 10.18%ã€21.36%ã€11.9% and 5.75% from 0.01 to 10 mg·L-1 with comparison respectively.0.1 mg·L-1 CRZ-1 could apparently increase the chlorophyll content in leaves of wheat by 15.79%(from the second day treatment) and it could apparently decrease the accumulation of MDA. At meanwhile, CRZ-1 could increase the activities of reactive oxygen scavenging enzyme, such as SOD and POD and decrease the accumulation of O2-. These results indicated that CRZ-1 was effective in promoting the growth of wheat under drought stress in a dose-dependent manner. Effect of 0.1 mg·L-1 CRZ-1 was the most significant. |
