| Rice is one of the most important food crop in China and many other countries, whose yield is affected by many factors. Rice leaf as the significant organ of photosynthesis,its color affected rice’s production largely. Besides, the mutant of leaf color is important to study genomics and plant physiology, and it also be used as a marker character in crop breeding.Use of Agrobacterium transformation method, ZH11 as receptor, T-DNA insertion mutant library, which can find a variety of mutants representative. In this experiment, we get a faden yellow leaf mutant fyl1 as material and study a gene affecting rice leaf’s color by its functional verification.1.Phenotype by field observation, to find yellow leaf mutant fyl1. Field survey found that the gene are caused by a recessive gene, and mutant fyl1’s height, tiller, per panicle were less than wild-type, especially the per panicle,mutant basically does not bear seeds.2.Through TAIL-PCR, we got the flanking sequence of T–DNA insertion, sequence analysis showed that if the T-DNA is inserted between the fifth exon and the sixth exon, it cause gene transcription irregularly which can lead to mutation phenotype. Therefore, building the FYL1 gene’s overexpression vector and the RNAi vector to verify its gene function.3.Through to the transgenic rice phenotypic observation and RT-PCR validation that FYL1 gene is indeed a rice leaf color control related new genes. |
