This research through to the key technology of Plumbago auriculatain large-scale production of pollen and seed collection and storage technology research, in order to improve the pollen quality and quantity of Plumbago auriculatain which can overcome the types by the Plumbago auriculatain alien lead to the difficulty of low natural setting in, lay the foundation for production of high quality seed, break the seed of Plumbago auriculatain rely on foreign imports of a bad situation.The results are as follows:1、 By comparing the Plumbago auriculatain pollen viability of different forms of openning, the pollen of Plumbago auriculatain in the semi open period, flower loose expansion, showing half spread shape recovery, the highest activity, L-type pollen viability was 84.53±0.32%, S type pollen viability was 86.26±0.17%, followed by the full flowering stage, bud stage picking the lowest pollen vitality.2、Under the condition of dry of 25℃, the pollen viability of Plumbago auriculatain under different drying time after briefly stored each are not identical,L-type pollen dry 0.5h, the most active for after low temperature storage,S-type Pollen is dry 1 h,most active after low temperature storage with the extension of drying time, the Plumbago auriculatain both type of pollen vigor drops rapidly, probably caused by dehydration.3、Through the ultra low temperature storage of the pollen of Plumbago auriculatain pre freezing treatment, the freezing effect of L type and S type pollen are:not precool liquid nitrogen frozen>20℃ precool 1h>0℃ precool 1h>4℃ precool 1h,the freezing and thawing effect is not ideal; the effect of L type and S type pollen storage:the water wash bath>40℃>0℃>ice bath room temperature of 25℃,to sum up, Lan Huadan, in the ultra low temperature storage is not needed before freezing, thawing is best way to tap water.4、Contrast of Plumbago auriculatain pollen right after drying and Plumbago auriculatain pollen without drying,under the condition of the same storage temperature, the dry pollen vitality of Plumbago auriculatain falling trend over time significantly lower than the fresh pollen of Plumbago auriculatain;And under different storage temperature, the lower the temperature, the pollen vitality of Plumbago auriculatain decline slower, its effect for the storage:-86℃>-20℃>4℃>25℃, so-86℃ cryogenic storage after drying Plumbago auriculatain for the optimal storage. After 50 d, the pollen vitality of L-type is still as high as52.31±0.30%, the pollen vitality of S-type is 52.81±0.24%, can be used for artificial pollination in the production.5、According to the "seed length and seed coat color" standard will be the seed of the Plumbago auriculatain is divided into 9 levels, through comparing with the germination rate of the seed coat:when the lengths are the same, the deeper the color, the higher the seed vigor. In addition to the green seed, the same color, length, seed germination rate is higher, the 100 seed weight and width index decreased gradually along with the seed maturity. When d≧13 mm, dark brown seeds germination rate was 83.33±2.72%,11 mm>d>13 mm, dark brown seeds germination rate was 73.33±2.72%, both of which are the top seed, can be used as a guidance standard picking the quality seed production of Plumbago auriculatain.6、Using different drying methods on Plumbago auriculatain seeds drying, vitality after cryogenic storage:oven 30℃>indoor air-dried ld>silica gel desiccant is drying1 d>oven 40℃>oven to 50℃, thus obtained the best dry Plumbago auriculatain seed in 30℃oven drying until natural air-dry condition. The seed moisture content is out of step with the germination rate, but to a certain extent affect the seed storage.7Contrast properly dry Plumbago auriculatain seed with fresh Plumbago auriculatain seed,under the same storage temperature, fresh seeds fall of energy over time significantly greater than dry seed vitality. Storage temperature is lower, after drying the Plumbago auriculatain seed vitality decline more slowly. Therefore,-86℃ cryogenic storage effect is best, seed remains more than 50% after 3 months of activity;While fresh Plumbago auriculatain seed does not like below 0℃ low temperature storage, at 20 ℃ and 86 ℃ temperature storage effect is not ideal, storage at 4℃ temperature effect is best. This may be due to fresh Plumbago auriculatain seed moisture content is higher,under 0℃ low temperature easily inside cells or between cells to produce cold freezes. So the best way to store Plumbago auriculatain seed is 30℃ oven drying until natural air-dry condition in cryogenic storage-86℃.
