According to the laboratory identification of duck plague virus (DPV) UL10 and UL48 gene, carried out plague virus in infected duck embryo fibroblasts, immunofluorescence techniques UL10 and UL48 gene expression product distribution in the cell characteristics testing. Obtain the following results:1、the cellular localization of the protein UL10The results show specific fluorescence may be detected after 4h plague virus infection in the cytoplasm, the nucleus fluorescence in a small amount of memory, with the time after infection, the specific fluorescence in the cytoplasm appears to be gradually increased in later stages of infection fluorescent membrane gradually goes after infection 48h, the amount of UL10 gene expression reached a maximum, the specific maximum fluorescence after infection 72h, apparent specific fluorescence began weakened portion of the shrinkage has occurred nucleus, cytoplasm appeared broken.2、the cellular localization of UL48 proteinThe results show specific fluorescence may be detected after 1h plague virus infection in the cytoplasm, with time after infection, there is more number of specific fluorescence in cytoplasm, and as time goes on, the more fluorescence gradually goes to the nuclear membrane, after infection 9h, UL48 gene expression reached a maximum amount, can be observed in the cytoplasm of a large number of fluorescent substances, at 24h, specific fluorescence decreased significantly after the start of infection after infection 72h, memory cells in the amount of fluorescence has rarely, some nuclear shrinkage and fragmentation has occurred, there is a clear cytoplasmic vacuoles.
